Kinetics of the lethal effect of actinomycin D on normal and leukemic cells.

چکیده
Spleen colony assays were used to quantitate the cytocidal effect of actinomycin D on leukemic cells and normal hematopoietic stem cells. Different doses of actinomycin D were administered to either normal or leukemia-bearing mice, and the surviving fraction of these cell populations was determined. Following single injections of the drug, the killing of both the normal and leukemic cell popula tions was found to occur over an extended period of time. The kinetics of killing of both cell populations was dose dependent with regard to both the final survival level and the time at which the minimum survival level was reached. The malignant cells exhibited a greater sensitivity than the normal cells. Both this difference in sensitivity and the ki netics of the lethal action of actinomycin D on both cell populations are discussed in terms of the difference in the proliferative states of the two cell populations and the known cellular effects of actinomycin D.

متن کاملAnti-Oxidative and Anti-Apoptotic Effects of Apigenin on Number of Viable and Apoptotic Blastomeres, Zona Pellucida Thickness and Hatching Rate of Mouse Embryos
Objective Apigenin is a plant-derived compound belong to the flavonoids category and has the protective effects on different cells. The aim was to evaluate the effect of apigenin on the number of viable and apoptotic blastomeres, the zona pellucida (ZP) thickness and the hatching rate of preimplantation mouse embryos exposed to H2O2 and actinomycin D. MaterialsAndMethods 420 two-cell embryos we...
متن کاملSperm DNA damage in mice irradiated with various doses of X-rays alone or in combination with actinomycin D or bleomycin sulfate: an in vivo study
Background: DNA damage in male germ cells due to exposure to environmental and manmade physico-chemical genotoxic agents is considered as the main cause of male infertility. The aim of this study was to evaluate the effects of combined modalities (radiotherapy and chemotherapy) routinely used for cancer treatment on mouse sperm chromatin in vivo. Materials and Methods: Forty-eight mice were div...
متن کاملEFFECTS OF SERUM AND PROTEIN SYNTHESIS INHIBITORS ON POLYMORPHONUCLEAR NEUTROPHIL APOPTOSIS
In this study the programmed cell death (apoptosis) of human neutrophilic granulocytes was investigated in the presence and absence of fetal calf serum (FCS), cycloheximide and actinomycin-D. The results show that when FCS is omitted from cultures, apart from a decrease in viability, the percentage of apoptotic cells and DNA fragmentation increases. Apoptosis is accelerated in serum withdra...
متن کاملCytotoxic effect of Pregabalin on U937 and Molt-4 Leukemic Cells in Vitro
Background: Pregabalin, a selective inhibitor of voltage dependent calcium channels has been used for treatment of epilepsy, fibromyalgia, generalized anxiety disorder and especially neuropathic pains. The anti-inflammatory properties of pregabalin in some pathologic conditions like orofacial pain and streptozotocin-induced diabetic mice have been reported. Effectiveness of pregabalin in treatm...
